I'm curious how that works. Tyrion demanded trial by combat and so they had to honor it ... even kings must honor that, right? Can the Faith override or deny these things? I'm guessing there's some "fine print" somewhere that the High Sparrow will use to his advantage. He's a very crafty old fellow! I suspect they won't deny the right to Trial by Combat but just won't allow Gregor to be Cersei's champion (which is all they would care about). They could pull out a number of arbitrary reasons (such as a KG not being eligible to battle against his own King's side or that Gregor isn't really "alive" so not a candidate to be a champion etc). No one knows for certain if Gregor is alive or not and since King's Landing apparently doesn't know about Beric, or Jon - they wouldn't know much about the resurrection thing - or at least it has not been mentioned anywhere in court or otherwise. They would have to assume he's a living person even though it's fairly obvious from what little you can see under his helm that something's not quite right. I could definitely see the Kingsguard being used as a rule. A technicality really. If Tommen still considers Gregor part of the kingsguard then he can order him to stand down and not fight for Cersei.

Maybe HS will arrange situation which provoke Gregor to use violence so that he'll be charged with murder and because this accusation will be excluded from the opportunities of being Cersei's champion in trial by combat. Or whatever, tbh every reason will be good enough for HS This is not a bad shout IMO. We know "I choose violence" is in this episode, so if Gregor goes nuts and flattens a bunch of Sparrows in the Red Keep, would the faith allow him to stand as Cersei's champion?

And Eugene Simon pointed in an interview to pay attention who attacks first. But my prediction even in such minor matters on the event wasn' confirmed by now, and so here I'll be careful. Countless scenarios are likely, since they not doing a lot of bending reality, characters, etc. for moving on the story.
